We previously observed that whenAzotobacter vinelandii was transformed by different broad-host-range plasmids, normal cellular functions such as growth and siderophore production are impaired. In the present work, whenA. vinelandii was transformed with the low copy number plasmid pRK290, the extent of this metabolic impairment was lessened, as evidenced by increased siderophore production and moderate levels of growth on medium that lacks added iron. It is concluded that the severity of the plasmid-induced metabolic load reflects the relative level of expression of plasmid-encoded proteins.  相似文献

Opiate receptors have been identified within the striatum and some have been localized presynaptically to nigrostriatal neurons. Using unilateral ablative lesions of the substantia nigra, we examined binding in the ipsilateral and contralateral striata. Lesions significantly lowered both 3H[D-Ala2,MePhe4,Gly(ol)5]enkephalin (DAGO) and 3H[D-Ala2,Leu5]enkephalin (DADL) binding. The inclusion of competitors in these assays revealed a decrease in both mu1 and mu2 receptors. Mu1 binding was slightly more sensitive to the lesioning than mu2 binding. Selective mu1 and mu2 binding assays supported these observations. No change in delta binding was observed in the lesioned striata. These studies raise the possibility that both mu1 and mu2, but not delta, receptors are localized presynaptically on nigrostriatal neurons.  相似文献

1. Phospholipids prelabelled with [(14)C]acetate, [(32)P]phosphate, [(3)H]- or [(14)C]-choline or [(3)H]inositol are not significantly degraded during fusion of Lettrée cells mediated by Sendai virus, nor are carbohydrates prelabelled with [(3)H]fucose, [(14)C]galactose or [(3)H]glucosamine. Less than 1nmol of lysophosphatidylcholine/10(7) cells is formed during fusion. Diethyl p-nitrophenyl phosphate, which inhibits phospholipase A by more than 95% has no effect on fusion. It is concluded that none of the events leading to cell fusion is accompanied by significant turnover of phospholipids or other membrane components. 2. Intracellular K(+) leaks out during virally mediated cell fusion; the loss is not as extensive as that of intracellularly accumulated choline or deoxyglucose. Movement of Ca(2+) into or out of cells could not be detected. 3. At concentrations of Lettrée cells insufficient to be agglutinated by virus, intracellularly accumulated choline and deoxyglucose leak out. Agglutination caused by concanavalin A does not result in leakage of intracellular metabolites. 4. P815Y cells, which agglutinate but do not fuse in the presence of virus, show leakage of intracellularly accumulated metabolites. The extent of leakage does not alter during the G(1) and S periods of the cell cycle. 5. Leakage is inhibited by Ca(2+), but is unaffected by EDTA. 6. It is concluded that the interaction of Sendai virus with mammalian cells causes a weakening of membrane integrity so that intracellular metabolites leak out. Such destabilization may facilitate viral entry and is therefore an interesting system for further biochemical studies.  相似文献

Approximately 75% or more of the L2 and L3 juvenile stages of the free-living nematode Caenorhabditis briggsae survived freezing and thawing without loss of fertility. Optimum survival depended upon a combination of conditions: (1) pretreatment with 5% DMSO at 0 °C for 10 min, (2) 0.2 °C per minute cooling rate from 0 to ?100 °C prior to immersion into liquid nitrogen, and (3) a 27.6 °C per minute warming rate from ?196 °C to ?10 °C. Storage at ?196 °C for more than 100 days was without effect on viability or fertility. Some of the L4 (about 50%) and adult (about 3%) stages survive the routine freeze-thaw treatment. However, there was no recovery of either embryonic stages or embryonated eggs from ?196 °C under these standard conditions. Either very fast cooling (about 545 °C/min) or fast warming (about 858 °C/min) rates diminished survival of the L2 and L3 stages drastically.Scanning electron microscopy revealed that freeze-thaw survivors with aberrant swimming behavior had cuticular defects. In juvenile forms, the altered swimming motion was lost after a molt whereas as abnormal adults grew, sinusoidal movement resumed. In the L4 and adult forms the cuticular abnormalities lowered viability and fertility. It is concluded that survival of nematodes from a freeze-thaw cycle is contingent upon establishing specific cryobiological conditions by varying aspects of the procedure that gave high recoveries of L2 and L3 stages.  相似文献

Aim

We documented how the similarity of mammal assemblages on continental and oceanic islands has changed since initial human colonization, since European arrival and overall. We investigated how levels of similarity might change in the future.

Methods

We used mammal occurrence data on islands to calculate the change in similarity using a pairwise approach based on Jaccard's index and a multisite approach based on Jaccard's and Sørensen's measures. We divided the mammal assemblages into two time periods, before and after island colonization or trade began with Europeans. We unpacked the mechanisms driving changes in similarity, exploring how initial similarity interacts with seven types of species turnover events to determine overall change. Finally, we calculated how future similarity levels will change if past trends in introductions and extinctions continue.

Results

Mammals, on both continental and oceanic islands, show one of the most pronounced cases of homogenization ever observed, and on oceanic islands mammals show the largest increase in homogenization ever observed for a terrestrial group. Most of the homogenization observed to date has been driven by recent historical changes, not by changes that occurred before European arrival. If current patterns of species introductions and extinctions continue, then oceanic islands will experience little additional homogenization, whereas continental islands will homogenize greatly beyond current levels.

Main conclusions

Mammal assemblages on oceanic islands show nearly an order of magnitude greater change in similarity than plant and bird assemblages. Projections of future similarity indicate that continental and oceanic islands are on different trajectories of change. These trajectories could be altered by management actions, but in some cases those actions that would be impactful run counter to current conservation norms.  相似文献

The habitat distribution of overwintered eggs, which were found to be the only source of spring recruitment of Argulus coregoni Thorell, was studied at a commercial fish farm in Central Finland. The frequency of occurrence of egg clutches in the deep parts of the canals and ponds was 50 to 80% and the percentage cover of the surface of stones with egg clutches was 1.7 to 6.4%, while in the shallow parts these values were 8 to 27% and 0.1 to 0.3%, respectively. A greater proportion of empty egg-shells was observed in shallow water in the mid-May, suggesting an earlier hatching there stimulated by the increased temperature and higher illumination. Under laboratory conditions, only elevated UV illumination, but not diurnally fluctuating temperature, significantly accelerated hatching. Normally overwintered eggs produced a pronounced peak of hatched larvae at the end of May and hatching continued at a much slower rate throughout the summer. Eggs that overwintered twice, first normally and then for a second time buried under sediments, were exposed to the same laboratory conditions simultaneously with normally overwintered eggs, but their hatching was delayed until August. The hatching rate was low, but markedly increased in December.  相似文献
